HLA IHBT Department introduces modernized sequencing technologies for searching blood-forming cell donors

As of 20 July 2026, the HLA Department IHBT introduced updated HLA typing procedures utilizing next-generation sequencing (NGS) methods within the framework of the approved flexible scope of accreditation (FRA).

These changes primarily concern the processing of samples from patients and potential donors in the context of searching for suitable hematopoietic stem cell donors for transplantation, where rapid and accurate HLA typing is a key prerequisite for selecting the optimal donor.

The modernization includes two significant technological improvements:

  • Extension of urgent HLA typing capabilities using Oxford Nanopore technology (MinION) – the previously used procedure designed primarily for rapid processing of individual urgent samples has been expanded to allow simultaneous processing of a larger number of samples in a single analytical run. This enables more flexible response to the needs of transplant centers, especially in cases of time-sensitive suitable donor searches.
  • Introduction of a new generation of standard NGS sequencing on the MiSeq i100 platform (Illumina) – the new sequencer replaces the previously used MiSeq instrument and provides higher data quality, greater operational efficiency and shorter laboratory processing time while maintaining high analytical accuracy.

All new procedures were properly verified and validated prior to implementation in accordance with the requirements of the ČSN EN ISO 15189 standard. Their goal is to further improve the quality, reliability and accessibility of laboratory services provided to patients, transplant centers and other clinical facilities.

Nothing changes for those ordering tests – the range of available tests, clinical indications, ordering methods and the format of result reports remain unchanged. The introduced innovations represent technical improvements to laboratory procedures that contribute to more efficient and faster sample processing.
