Department of Biochemistry
The workplace is incorporated into the Complement of laboratories IHBT. It continuously defends an independent and impartial assessment of professional competence of the Complement of laboratories according to the standard ČSN EN ISO 15189, whose application is verified by the Czech Accreditation Institute. Examination methods within the scope of accreditation are defined by the attachment of the Accreditation Certificate in the current version, which is available on the website IHBT.
The Department of Biochemistry has long been engaged in research into changes in the composition and function of low-molecular and high-molecular substances contained in blood, which occur in onco-hematological diseases, hemostasis disorders and thrombosis, for example in the context of cardiovascular diseases.
We monitor the relationships between individual components of the coagulation cascade, from monitoring the activities of serine proteases involved in the formation of fibrin network, the structure of the formed network, to fibrinolysis. We detect both acquired and hereditary mutations of fibrinogen and seek the relationship between altered structure and its properties. Methodologically, we use new approaches to study diseases in the form of combining liquid chromatography with mass spectrometry (LC-MS) for metabolomics and proteomics. We also use 3D imaging, monitoring of interactions in real time and theoretical methods of structural biology, such as molecular modeling and molecular dynamics. The aim of our research work is to clarify physiological and pathophysiological processes in selected diseases and to further use these findings in diagnostics or in monitoring treatment.
Within routine examinations, the department offers spectrophotometric determination of free hemoglobin in plasma (accredited method), SRA test for the determination of heparin-induced thrombocytopenia (HIT), genetic and functional examination of fibrinogen, determination of malondialdehyde (MDA), and monitoring of drug levels (TDM, Therapeutic Drug Monitoring) by LC-MS/MS method for the determination of 8-methoxypsoralen (8-MOP), busulfan, tyrosine kinase inhibitors (TKI), and new methods within the scope of accreditation for the determination of antifungal agents (posaconazole, voriconazole, isavuconazole) and immunosuppressants (cyclosporine A, tacrolimus, sirolimus).
