Department of Biochemistry
The workplace is included in the Complement of laboratories at ÚHKT. It continuously defends an independent and impartial assessment of the professional competence of the Complement of laboratories according to the standard ČSN EN ISO 15189, whose application is verified by the Czech Institute for Accreditation. Examination methods within the scope of accreditation are defined by the appendix of the Accreditation Certificate in the current version, which is available on the website ÚHKT.
The Department of Biochemistry has long been engaged in research into changes in the composition and function of low-molecular and high-molecular substances contained in blood, which occur in oncohematological diseases, in hemostasis disorders and in thrombosis, for example in the context of cardiovascular diseases.
We monitor the relationships between individual components of the coagulation cascade, from monitoring the activities of serine proteases involved in the formation of fibrin networks, the structure of the formed network, to fibrinolysis. We identify acquired and inherited mutations of fibrinogen and look for relationships between the altered structure and its properties. Methodologically, we use new approaches to studying diseases in the form of combining liquid chromatography with mass spectrometry (LC-MS) for metabolomics and proteomics. We also use 3D imaging, real-time interaction monitoring, and theoretical methods of structural biology, such as molecular modeling and molecular dynamics. The aim of our research work is to clarify physiological and pathophysiological processes in selected diseases and further use these findings in diagnosis or in monitoring treatment.
Within routine examinations, the department offers spectrophotometric determination of free hemoglobin in plasma (accredited method), SRA test for determination of heparin-induced thrombocytopenia (HIT), genetic and functional fibrinogen examination, determination of malondialdehyde (MDA), and drug level monitoring (TDM, Therapeutic Drug Monitoring) using LC-MS/MS method for determination of 8-methoxypsoralen (8-MOP), busulfan, tyrosine kinase inhibitors (TKI), and new methods within the scope of accreditation for determination of antifungals (posaconazole, voriconazole, isavuconazole) and immunosuppressants (cyclosporin A, tacrolimus, sirolimus).
