Hematopoietic Cell Interaction with the Proteins of the Extracellular Matrix

Aside from retaining stem and progenitor hematopoietic cells in the bone marrow (BM), engagement of adhesion receptors on these cells plays an important role in the regulation of hematopoiesis. Some diseases of the hematopoietic system are characterized by aberrant cell adhesiveness that result in abnormal regulation of growth, differentiation, or survival of hematopoietic cells. Also, leukemic cell adhesion to the bone marrow microenvironment often confers resistance to chemotherapy drugs, promotes leukemia cell survival and contributes to the residual disease.

However, the nature of adhesion structures formed by immature forms of hematopoietic cells is not well explored. The basic molecular mechanisms governing the assembly and dynamics of adhesion structures in hematopoietic cells are rather inferred from the knowledge obtained in adherent cell models.

We have developed methods for the analysis of hematopoietic cell interactions with proteins of extracellular matrix (real-time microimpedance measurement, interference reflexion microscopy) and we combine them with the analysis of selected proteins, such as Src family kinases or small Rho GTPases and their effectors (grant project No 16-16169S from the Grant Agency of the Czech Republic) to characterize the major signalling pathways regulating these interactions.
